چکیده

Abstract Background Prediabetes is characterized by a hemoglobin A1c of 5.7–6.4% and fasting blood glucose 100–125 mg/dl. A high percentage prediabetes subjects develop type 2 diabetes mellitus in the next years. The effects opioid peptides their receptors, addition to immunological cytokines, on are not well understood. Therefore, molecular, physiological, clinical studies required link system, immune insulin resistance (IR) prediabetes. We hypothesize that (endomorphin-2 (EM2), β-endorphin (βEP)), receptors (µ-opioid (MOR) κ-opioid (KOR)), inflammatory cytokines (IL-6) anti-inflammatory cytokine (IL-10), affect IR parameters patients with Methods Sixty (prediabetes+IR) sixty without (prediabetes-IR), 58 controls, have participated study. IL-6, IL-10, EM2, βEP, MOR, KOR were measured ELISA technique. Results In general, most dyslipidemia. β-endorphin, endomorphin-2 higher subgroups than control group. system was activated an IR-dependent manner. Prediabetes+IR can be predicted increased levels EM2 combination IL-10 EM2/KOR good sensitivity specificity. Conclusion Opioid upregulated prediabetes, depending significance cytokines. intercorrelation between EOS, confirmed.

Pharmacological profiles of opioid ligands at Kappa opioid receptors

BACKGROUND The aim of the present study was to describe the activity of a set of opioid drugs, including partial agonists, in a human embryonic kidney cell system stably expressing only the mouse kappa-opioid receptors. Receptor activation was assessed by measuring the inhibition of cyclic adenosine mono phosphate (cAMP) production stimulated by 5 microM forskolin. Intrinsic activities and pote...

Serum vitamin-D predicts insulin resistance in individuals with prediabetes

BACKGROUND & OBJECTIVES Patients with diabetes and vitamin-D insufficiency have increased insulin resistance. Similar observations among individuals with prediabetes are not well documented. The aim of this study was to find the occurrence of vitamin-D insufficiency/deficiency among individuals with prediabetes and to evaluate the relationship between vitamin-D status and insulin resistance. ...

Homeostasis Model Assessment of Insulin Resistance (HOMA-IR) in the Diagnosis of Insulin Resistance and Prediabetes

Type 2 diabetes mellitus consists of metabolic disturbances characterised mainly characterized by hyperglycemia which usually result from the combination of resistance to insulin action, inadequate insulin secretion, and excessive or inappropriate glucagon secretion. In clinical practice its very difficult to establish how much part of this metabolic disturbances is due to lack of insulin and h...

Endomorphin-2: a biased agonist at the μ-opioid receptor.

Previously we correlated the efficacy for G protein activation with that for arrestin recruitment for a number of agonists at the μ-opioid receptor (MOPr) stably expressed in HEK293 cells. We suggested that the endomorphins (endomorphin-1 and -2) might be biased toward arrestin recruitment. In the present study, we investigated this phenomenon in more detail for endomorphin-2, using endogenous ...

A novel fasting blood test for insulin resistance and prediabetes.

BACKGROUND Insulin resistance (IR) can precede the dysglycemic states of prediabetes and type 2 diabetes mellitus (T2DM) by a number of years and is an early marker of risk for metabolic and cardiovascular disease. There is an unmet need for a simple method to measure IR that can be used for routine screening, prospective study, risk assessment, and therapeutic monitoring. We have reported seve...
